Genomic DNA Extraction Protocol

Materials Required:

  • HyperSilMag Pure DNA Kit
  • Biological sample (blood, tissue, cultured cells)
  • Lysis Buffer (provided)
  • Binding Buffer (provided)
  • Wash Buffer (provided)
  • Elution Buffer (provided)
  • Magnetic separator

Procedure:

  1. Sample Lysis: Add 200 uL sample to 200 uL Lysis Buffer. Incubate at 56 C for 10 minutes.
  2. Binding: Add 200 uL Binding Buffer and 20 uL magnetic beads. Mix well and incubate for 5 minutes at room temperature.
  3. Magnetic Separation: Place tube on magnetic separator for 2 minutes. Remove supernatant.
  4. Washing: Add 500 uL Wash Buffer. Mix gently, separate magnetically, and discard supernatant. Repeat once.
  5. Drying: Leave tube on magnetic separator for 5 minutes to air-dry beads.
  6. Elution: Add 50-100 uL Elution Buffer. Incubate at 56 C for 5 minutes. Separate magnetically and transfer DNA solution to clean tube.

Yield: Typically 5-20 ug DNA from 200 uL blood sample.

Purity: A260/A280 ratio of 1.8-2.0

Time: Total protocol time approximately 30 minutes
