Protein Coupling Protocol

Materials Required:

  • HyperSilMag Carboxyl
  • Target protein or antibody
  • EDC (1-Ethyl-3-(3-dimethylaminopropyl)carbodiimide)
  • NHS (N-Hydroxysuccinimide)
  • MES Buffer (pH 5.0-6.0)
  • PBS Buffer (pH 7.4)
  • Blocking Buffer (1% BSA in PBS)

Procedure:

  1. Bead Preparation: Wash beads 3x with MES buffer using magnetic separation.
  2. Activation: Resuspend beads in MES buffer containing EDC (2 mg/mL) and NHS (5 mg/mL). Incubate for 15 minutes at room temperature.
  3. Washing: Remove activation solution and wash beads 2x with cold PBS.
  4. Coupling: Add target protein (0.1-1 mg/mL) in PBS. Incubate 2 hours at room temperature with gentle rotation.
  5. Blocking: Remove unbound protein and add blocking buffer. Incubate 30 minutes.
  6. Final Wash: Wash 3x with PBS. Store at 4 C in PBS with 0.02% sodium azide.

Yield: Typically > 80% coupling efficiency for most proteins.
