Materials Required:
- HyperSilMag Streptavidin
- Biotinylated target molecule (protein, nucleic acid, etc.)
- Binding Buffer (PBS with 0.05% Tween-20)
- Wash Buffer (PBS with 0.1% Tween-20)
- Elution Buffer (optional, for recovery)
- Magnetic separator
Procedure:
- Bead Preparation: Wash streptavidin beads 2x with Binding Buffer using magnetic separation.
- Binding: Resuspend beads in Binding Buffer. Add biotinylated target (0.1-10 ug/mL). Incubate 15-30 minutes at room temperature with gentle rotation.
- Washing: Magnetically separate and wash 3x with Wash Buffer to remove unbound material.
- Application: Use beads directly for assays, or proceed to elution if target recovery is needed.
- Elution (Optional): Add Elution Buffer (e.g., biotin solution 5 mM). Incubate 10 minutes with mixing. Magnetically separate and collect eluate.
Binding Capacity: >3000 pmol biotin per mg beads
Affinity: Kd = 10^-15 M (one of the strongest non-covalent interactions)
